Announcing a new article publication for
BIO Integration journal. Radiation therapy (RT) has a critical role in cancer treatment, yet the efficacy is often limited by tumor resistance mechanisms, such as cellular DNA repair activation, heterogeneous cell cycle, hypoxia, and an immunosuppressive microenvironment. Conventional radiosensitization strategies face significant challenges, which are caused by insufficient efficiency, a short treatment window, and off-target toxicities. In contrast, living cells offer a novel strategy to overcome these limitations by leveraging innate characteristics, including tumor chemotaxis and radiosensitization factor secretion. Living cells have recently been applied in radiotherapy to amplify tumor cell killing effects and avoid healthy tissue damage.
This article systematically summarizes recent advances in living cell-based radiosensitization strategies, emphasizing the dual roles as tumor targeting carriers and dynamic microenvironment modulators. The radiosensitization mechanisms underlying diverse cell types are analyzed. For example, stem cells enhance radiotherapy via STAT3-mediated DNA repair inhibition, while immune cells and bacteria use immunogenic cell death synergy to induce immune activation. The challenges and prospects of living cells in achieving radiosensitization are also highlighted.
