科学家发现保护生殖遗传物质关键机制


西班牙塞维利亚大学的科研团队发现了一种在生殖细胞中维持DNA完整性的关键过程,为理解生物在卵子形成过程中如何避免遗传缺陷提供了新的线索。

在近期发表的研究中-该研究成果源自努里亚·费尔南德斯-费尔南德斯(Nuria Fernández-Fernández)的博士论文研究-,研究人员发现,在秀丽隐杆线虫(Caenorhabditis elegans)中,BRC-1/BRD-1蛋白复合物的磷酸化(这是一种化学修饰形式)—如何作为DNA损伤的“开关”发挥作用。这种激活机制使基因组能在减数分裂过程中得到有效修复,而减数分裂正是生殖细胞形成的生物过程。

当该机制失效时,DNA会出现严重断裂,损伤也会不断累积,染色体会发生碎裂,最终导致胚胎丧失活性。研究人员还发现,布鲁姆酶(Bloom)与Mus81酶这两种酶会因错误修复受损DNA分子间的连接而加剧了这些问题。

“我们的研究揭示了细胞应对生殖细胞基因损伤的关键环节”,安达卢西亚分子生物学与再生医学研究中心(Cabimer)首席研究员塔蒂亚娜·加西亚-缪斯(Tatiana García-Muse)博士指出:“理解这些分子过程,对于阐明如何防止非整倍体及发育缺陷至关重要。”
