Announcing a new article publication for
Zoonoses journal. Flaviviruses remain a formidable global health burden, necessitating therapeutic strategies that can overcome the inherent limitations and safety risks of traditional envelope-targeted approaches. Non-structural protein 1 (NS1) has emerged as a critical, multifunctional glycoprotein that orchestrates the viral life cycle and host pathogenesis. NS1 acts as a central hub for RNA replication intracellularly through essential interactions with the NS4A and NS4B proteins, while also regulating maturation of infectious progeny particles. Secreted NS1 (sNS1) functions as a potent virulence factor extracellularly that drives vascular leakage and immune evasion by directly disrupting the endothelial glycocalyx and antagonizing the complement system. Synthesizing current advances in NS1-targeted monoclonal antibodies has revealed a unique therapeutic capacity to circumvent antibody-dependent enhancement (ADE), while providing broad-spectrum protection across diverse flavivirus species. The synergistic mechanisms underlying sNS1 neutralization and Fc-mediated effector functions have been elucidated as a robust defense against established infection based on an analysis of representative antibodies, such as 1G5.3 and 2B7. Despite lingering translational hurdles regarding immune complex management, NS1-targeted strategies provide a safer and potentially transformative paradigm for the global prevention and control of flavivirus-associated diseases.
