Drug Development and Promising Therapeutic Targets for Coronary Artery Disease: Current Landscape and Future Prospects

Announcing a new article publication for Cardiovascular Innovations and Applications. Coronary artery disease (CAD) remains the leading cause of death globally, despite the limited efficacy of current pharmacological therapies. The need for safer and more effective treatment strategies has never been more urgent. This review provides a contemporary overview of advances in CAD drug development. Novel lipid-lowering agents such as proprotein convertase subtilisin/kexin type 9 (PCSK9) and angiopoietin-like protein 3 (ANGPTL3) inhibitors, anti-inflammatory therapies targeting interleukin pathways, and metabolic modulators have expanded the therapeutic landscape. Concurrently, precision medicine and drug repurposing approaches, informed by genomic profiling and biomarker-based stratification, are transforming the framework of CAD therapy. The integration of artificial intelligence into drug discovery further accelerates innovation. Herein, we summarize the current state of CAD pharmacotherapy, highlight recent translational and clinical breakthroughs, and discuss future directions aimed at addressing unmet needs and improving outcomes in this high-risk population.
